چکیده

Automated 3D Reconstruction of Neuronal Structures from Serial Sections. (August 1999) Brent P. Burton, B.S., Texas A&M University Chair of Advisory Committee: Dr. Bruce H. McCormick A fast automated system for tracing neurons in parallel is described, adequate to support a quantitative analysis of neuron morphology. The system described here automates digitized neuron feature extraction and reconstruction, thereby replacing current largely manual techniques for tracing individual neurons. Serial sections of brain tissue are created by physical sectioning. Sections are processed during scanning to determine regions of interest (ROIs) and to quickly cull unnecessary image data. An aggressive data culling and compression scheme reduces the original volumetric data into a ROI-based image collection that makes temporary secondary storage feasible. Neighboring ROIs are then matched from successive sections for segment and ber tracing. Reconstructed segments created from these matches are disambiguated, resulting in an abbreviated structural description of the tissue's neurons and ber tracts.

Important Factors for the Three-Dimensional Reconstruction of Neuronal Structures from Serial Ultrathin Sections

Quantitative analysis of anatomical synaptic connectivity in microcircuits depends upon accurate three-dimensional (3D) reconstructions of synaptic ultrastructure using electron microscopy of serial ultrathin sections. Here we address two pitfalls in current methodology that lead to inaccurate reconstructions and compromise conclusions drawn from the data. The first pitfall is inaccurate determ...

Three-Dimensional Characterization of Microstructure by Electron Back-Scatter Diffraction

The characterization of microstructures in three dimensions is reviewed, with an emphasis on the use of automated electron backscatter diffraction techniques. Both statistical reconstruction of polycrystalline structures from multiple cross sections and reconstruction from parallel, serial sections are discussed. In addition, statistical reconstruction of second-phase particle microstructures f...

Reconstructing and Visualizing Models of Neuronal Dendrites

Neuroscientists have studied the relationship between nerve cell morphology and function for over a century. To pursue these studies, they need accurate threedimensional models of nerve cells that facilitate detailed anatomical measurement and the identification of internal structures. Although serial transmission electron microscopy has been a source of such models since the mid 1960s, model r...

Understanding 3-Dimensional World from 2-Dimensional Immunofluorescent Adjacent Sections

In many fields of biological sciences including embryology and cancer research, understanding of 3-dimensional structures is crucial to uncovering normal and pathological phenomena. While the most optimal method would be to directly observe the complete object without any destruction, staining and imaging of thick sections and whole mount samples can be challenging. For decades, researchers hav...

3D reconstruction and characterization of polycrystalline microstructures using a FIB–SEM system

A novel methodology is described in this paper which is a step towards three-dimensional representation of grain structures for microstructure characterization and processing microstructural data for subsequent computational analysis. It facilitates evaluation of stereological parameters of grain structures from a series of two-dimensional (2D) electron backscatter diffraction (EBSD) maps. Crys...
